Summary: Tandem High-Dose Therapy with Melphalan and Total Marrow Irradiation (TMI) with Peripheral Blood Progenitor Cell Support and Dexamethasone and Thalidomide Maintenance in Multiple Myeloma: A Phase I/II Trial
You have been asked to participate in this research study because you have multiple myeloma (MM). The purpose of this study is to evaluate the feasibility (how easily the treatment can be given) and toxicities (side effects) of tandem (two) cycle ablative (destroying bone marrow) chemotherapy (drugs that kill cancer cells). The first cycle consists of high-dose chemotherapy with melphalan followed by peripheral blood stem cell transplant (PBSCT) (receiving back previously collected, immature blood cells). The second cycle consists of fractionated (divided doses) total marrow irradiation (TMI, radiation directed as much as possible to the bone marrow) followed by PBSCT. Another purpose of this study is to determine the highest dose of TMI that can be given without causing unmanageable side effects. Afterwards, Dexamethasone and Thalidomide will be given to help maintain any benefit obtained from the two cycles of high-dose treatment. Your participation in this study is expected to last approximately 3 to 6 months for the first two cycles of treatment, followed by up to 12 months of maintenance treatment. After treatment on this study is over, your medical condition will be followed indefinitely.
