A Phase I Trial of PS-341 in Combination with Topotecan in Advanced Solid Tumor Malignancies

You have been asked to participate in this research study because you have advanced cancer that is no longer responding to standard treatment. The purpose of this study is to find out the highest dose of topotecan that can be given in combination with a fixed dose of PS-341 without causing unmanageable side effects and to learn about the side effects of the treatment. An additional purpose is to study the level of topotecan in the body over time when given in combination with PS-341. A third purpose of this study is to evaluate how certain cells in a subject's body respond to topotecan and PS-341 after several weeks of treatment. Tissue used for this analysis will be collected from participants who volunteer to take part and have tumor tissue that is accessible for biopsy. Your treatment on this study is expected to last at least 12 weeks but may continue longer if your tumor is not getting bigger and you are not having unmanageable side effects. Once you complete treatment, your disease status will be followed indefinitely.
