The BeST Trial: A Randomized Phase II Study of VEGF, RAF kinase, and mTOR
The purpose of this study is to compare the effects, good and/or bad of several different treatments on metastatic renal cell carcinoma. This study is designed to determine which of the four drug regimens should be tested in later clinical trials. Four drug regimens experimental arms: Arm A (bevacizumab alone); Arm B (bevacizumab and temsirolimus); Arm C (bevacizumab and sorafenib); and Arm D (temsirolimus and sorafenib).
Bevacizumab is investigational and has not been approved by the FDA for use in this cancer. Sorafenib and temsirolimus have been approved by the FDA for the treatment of advanced renal cell carcinoma. However, when these two drugs are combined with each other or bevacizumab, their use is investigational.
Patient Inclusion Criteria:
- Renal (Kidney) Cancer with metastatic disease
- Age > 18 years
- Good performance status
- Prior nephrectomy is required
- Prior radiation is allowed
- No history of cancer in the last 5 years
Patient Exclusion Criteria:
- Clinical evidence of CNS disease, including primary brain tumor
- No clinically significant cardiovascular disease
Primary Outcomes: To access progression free survival of each treatment arm
