A Phase 1 Protocol of the Combination of Bortezomib and Tipifarnib for Relapsed or Refractory Multipe Myeloma

You have been asked to participate in this research study because you have multiple myeloma (MM) that has relapsed (returned after previous treatment) or refractory (has failed to respond to previous treatment).  The purpose of this study is to test the safety of taking bortezomib (VELCADE) and tipifarnib (Zarnestra™).  Another objective is to find the highest dose of each drug that can be given together without causing severe side effects.  Additional objectives are to evaluate the effectiveness of the combination in terms of changes in blood tests, overall response rate, how long response lasts, and length of survival without return of disease.  A series of blood samples will be drawn for pharmacokinetic (PK) testing that shows how the body processes the combination of study drugs over time.  Your treatment on this study is expected to last at least four weeks (one cycle of treatment).  If you are tolerating treatment and your medical condition is not getting worse, you will be able to receive additional cycles of treatment.  If you are benefiting from therapy, you may receive up to 8 four-week cycles of treatment.  After you discontinue treatment, your medical condition will continue to be followed indefinitely.
