Trial of Beta Blocker Therapy (Atenolol) vs. Angiotensin II Receptor Blocker Therapy (Losartan) in Individuals with Marfan Syndrome

The study is enrolling patients with Marfan syndrome to compare two drugs to see which one is better at slowing the rate of aortic root enlargement. The aorta is the large blood vessel that takes blood from the heart out to the body. Patients will receive either Atenolol or Losartan. The study will also examine which drug may have fewer side-effects.


Compensation Provided

Patient Inclusion/Exclusion Criteria:

You or your child may be eligible to participate if you or your child:
1)has the diagnosis of Marfan syndrome by Ghent criteria
2)are between 6 months to 25 years of age
3)have an enlarged Aortic root (Z-score > 3.0) A z-score is a calculation based on the size of the aorta compared to the weight and height of the person.

You or your child are not eligible for this study if you or your child:
1)have had aortic surgery or plan to have aortic surgery within 6 months of enrollment
2)require therapeutic use of angiotensin II receptor blocker or calcium channel blocker (e.g. for arrhythmia, ventricular dysfunction or valve regurgitation)
3)have intolerance to losartan or atenolol
4)have renal dysfunction
5)have severe asthma
6)have diabetes mellitus
7)are pregnant or plan to become pregnant within 3 years of enrollment
